Dr. Jin O-Uchi is interested in understanding the molecular mechanisms underlying sudden cardiac death and heart failure (HF) by ion channel disorders, specifically focusing on cellular and mitochondrial calcium (Ca2+)-handling disorders.

Current projects in the O-Uchi laboratory include investigations of 1) functional significance of adrenergic signaling for the regulation of mitochondrial Ca2+ influx mechanism in the cardiomyocytes; 2) the role of proline-rich tyrosine kinase 2 (Pyk2) in the HF development, especially focusing on the Pyk2-dependent phosphorylation of mitochondrial Ca2+ Uniporter (MCU) and 3) the pathophysiological role of mitochondria-localized ryanodine receptor type 1 (RyR1) in the heart using knock-in mice carrying a RyR1 mutation Y522S (YS mice) found in the human malignant hyperthermia (MH) family. Our central hypothesis is that chronic mitochondrial Ca2+ overload either via hyperphosphorylated MCU or leaky mutant RyR1 in cardiac mitochondria induces mitochondrial injury and cellular oxidative stress, which leads to HF and/or cardiac arrhythmia. His research projects is supported by American Heart Association (AHA) Beginning Grant-in-Aid, AHA Scientist Development Grant, Research Career Enhancement Award, American Physiological Society and Medical Research Grant from the W.W. Smith Charitable Trust.
